Product Introduction:
DNase I (Deoxyribonuclease I) is an endonuclease that can digest single-stranded or double-stranded DNA to produce monodeoxyribonucleotides or single-stranded/double-stranded oligodeoxyribonucleotides. The products of DNase I-catalyzed hydrolysis of single-stranded or double-stranded DNA have a phosphate group at the 5' end and a hydroxyl group at the 3' end. DNase I activity is dependent on calcium ions and can be activated by magnesium ions or divalent manganese ions. In the presence of magnesium ions, DNase I randomly cleaves double-stranded DNA at any site; in the presence of divalent manganese ions, DNase I cleaves both strands of DNA at the same site, generating blunt ends or sticky ends with 1-2 nucleotide overhangs.
Product Features:
After digestion with the company's proprietary DNase and unique reaction buffer, this product does not require tedious phenol/chloroform extraction for inactivation. It also eliminates the need to add EDTA before heat inactivation, avoiding the introduction of EDTA into RNA which may inhibit downstream reverse transcription reactions. It is extremely fast and convenient to use.
Application Scope:
Preparation of DNA-free RNA samples.Removal of potential DNA contamination such as genomic DNA from RNA samples before RT-PCR reactions.Removal of DNA templates after in vitro RNA transcription catalyzed by RNAPolymerases such as T7, T3, and SP6.DNase I footprinting to study DNA-protein interactions.Nick translation.Generation of random DNA fragment libraries.Partial cleavage of genomic DNA as a positive control in TUNEL assays for apoptosis detection.
Use 1 unit of DNase I per μg of RNA. If RNA is less than 1 μg, use 1 unit of DNase I. If RNA exceeds 1 μg, scale up the amounts of DNase I, 10× DNase Buffer, and reaction volume proportionally according to the RNA quantity.
