Product Introduction:
RNA is unstable and highly prone to degradation. Purified RNA dissolved in RNase-free TE buffer or water may degrade even when stored at -20°C. To solve this problem, RNA pellets or RNA solutions can be dissolved in RNA Long-term Storage Solution. This allows RNA to be stored at 4°C overnight or at -20°C for at least 1 year without degradation. RNA Long-term Storage Solution is the optimal choice for RNA sample transportation and medium-to-long-term storage. When needed, RNA can be recovered by conventional ethanol precipitation, or high-concentration RNA (up to 4 mg/mL) stored in the solution can be directly used for RNA electrophoresis and Northern Blot.
Dissolving RNA Pellets in RNA Long-term Storage Solution:
1) For Solid RNA Pellets:Add 1 μL of RNA Long-term Storage Solution per 0.4-4 μg of RNA pellet. Pipette repeatedly to mix or shake at room temperature for 15-30 minutes to dissolve the pellet.Dried RNA pellets are difficult to dissolve. After repeated pipetting, heat at 50°C for 10-15 minutes to assist dissolution.For better results, first dissolve the RNA pellet in a small volume of RNase-free TE buffer or water, then follow the operation steps for liquid RNA.
2) For Liquid RNA Solutions:Add 1 μL of RNA Long-term Storage Solution per 0.4-4 μg of RNA solution and mix well.Note that the volume percentage of RNA Long-term Storage Solution in the mixture should not be less than 80%.
3) Measure the OD value. Use the corresponding amount of RNA Long-term Storage Solution as a blank control.
4) Store the dissolved RNA sample at -20°C or -70°C.
Precipitating RNA from RNA Long-term Storage Solution:
1) Estimate the final volume of the RNA solution. Add 4 volumes of absolute ethanol and mix well.If the solution volume is too small for convenient operation, dilute the RNA solution with RNase-free water. If the RNA concentration in the solution is below 0.25 μg/μL, add RNase-free 5M NaCl to a final concentration of 0.2M, mix well, then add 4 volumes of ethanol.
2) Incubate at room temperature for 5 minutes.
3) Centrifuge at 12,000 rpm for 5 minutes. Discard the supernatant, air-dry the pellet, and redissolve it.
4) The redissolved RNA can be used for RT-PCR reactions and other experiments.
Direct Use of RNA in RNA Long-term Storage Solution:
Directly aspirate RNA from RNA Long-term Storage Solution for regular or formaldehyde-denaturing electrophoresis and Northern Blot. For formaldehyde-denaturing electrophoresis, the concentration of RNA Long-term Storage Solution in the final loaded sample can be as high as 50%.
Appendix: Preparation of Formaldehyde-Denaturing Electrophoresis Samples:Before use, mix water (87 μL), formaldehyde (81 μL), 50% glycerol containing 0.25 mg/mL bromophenol blue (48 μL), and 20× MOPS (24 μL).Mix the above mixture with an equal volume of the RNA sample in RNA Long-term Storage Solution.Incubate at 55°C for 10 minutes, then load the sample according to the standard formaldehyde-denaturing electrophoresis procedure.
