This kit utilizes DNA spin columns and a novel, unique buffer system, suitable for the rapid and simple extraction of genomic DNA from plant samples containing polyphenols, polysaccharides and enzyme inhibitors. DNA purification for one or more plant samples — 100 mg fresh or 20 mg dry tissue — can be completed within 30 minutes. The extraction procedure requires no toxic organic extractions such as phenol/chloroform, nor time-consuming isopropanol or ethanol precipitation. It can efficiently and rapidly remove impurities including polysaccharides, polyphenols and enzyme inhibitors. The purified DNA can be directly used in PCR, restriction enzyme digestion, hybridization and other experiments.

Fresh or dried plant tissues (cells) are ground and lysed with lysis buffer. Proteins, polysaccharides and cell debris are removed by precipitation. Genomic DNA then selectively binds to the silica membrane in the 96-well purification plate under high chaotropic salt conditions. Through a series of rapid wash-centrifuge steps, impurities such as polysaccharides, polyphenols, cellular metabolites and proteins are further removed. Finally, pure genomic DNA is eluted from the silica membrane with low-salt elution buffer.
